In this paper, we describe the discovery of a fully synthetic covalent ligand EN219 that mimics the action of the more complex natural product nimbolide that targets the E3 ligase RNF114 and we demonstrate that EN219 can be used as an RNF114 recruiter in targeted protein degradation applications.
